PROMETHEUS Study

PRecision gerOMedicinE: Tailored Healthy agEing with lifestyle, sUpplements and drugS

Study Overview

As populations age, extending healthspan—preserving physical, cognitive, and immune function—has become a public health priority. This 8-week, single-arm feasibility study at the NUS Academy for Healthy Longevity tests a precision geromedicine intervention tailored to adults aged 50–80.

Twenty participants will undergo a multimodal program of sleep and dietary guidance, supervised exercise with exergaming, motivational coaching, and supplementation with protein, creatine, and fucoidan. Participants with lower muscle, fitness, or cognitive scores will receive additional urolithin A, NMN, and/or multivitamins, with mid-study adjustments such as ergothioneine added based on response.

Primary outcomes include immune function, cognition, and muscle strength; secondary outcomes include biological age, sleep/activity patterns, and feasibility measures. Results will guide a 12-month trial for the XPRIZE Healthspan competition, combining lifestyle, nutraceutical, and repurposed drug strategies to slow aging.
